PURPOSE: To advantageously obtain the subject amylase useful for production of maltotriose by culturing microorganism belonging to genus Microbacterium and producing a maltotriiose-producing amylase and collecting said enzyme.
CONSTITUTION: A microorganism (e.g. Microbacterium sp. AM9581) belonging to genus Microbacterium and producing maltotriose-producing amylase is cultured. Next. maltotriose-producing amylase is collected. Resultant maltotriose- ptoducing amylase has an action producing mainly maltotriose by acting on starch and has substrate specificity affecting to amylose, amylopectin, glycogen and starch and not affecting to pullulan. Besides, optimum pH is about 6.5 in a reaction at 40°C for 30min, optimum temperature is about 50°C in a reaction at pH 7 for 30min, heat stability is improved by calcium ion and molecular weight by gel filtration method is about 40000.
