PURPOSE: To phosphorylate a hydroxyl group of nucleotide, etc. easily and selectively, and to obtain the titled compound such as nucleotide, etc. economically, by treating a hydroxyl group-containing compound with an Al hydride compound, reacting the resulting compound with a phosphorylating agent.
CONSTITUTION: First, a hydroxyl group-containing compound (preferably nucleotide) is treated with an aluminating agent [preferably (CH3)3Al, etc.] consisting of an Al hydride compound or an trialkyl Al compound. Then, a prepared aluminum alkoxide is reacted with a phosphorylating agent [preferably (C2H5O)2 POCl, etc.], to give the desired compound. The two reactions are preferably carried out in a solvent such as THF, etc. at 15W25°C for 1W2hr of total reaction time of the two reactions.
