PURPOSE: To obtain a polysaccharide having remarkably strong antitumor activity compares with known β-1,3-glucan and little difference in physical properties measured by analysis, by extracting FUKUROTAKE volvariella volvacea with an alkaline aqueous solution and extracting the extracted liquid further with an alkaline phenol.
CONSTITUTION: FUKUROTAKE is cut to small pieces and foreign materials are removed with a solvent comprising a combination of water, alcohol, etc. the purified product is extracted with an alkali at ≤30°C using 5W100 times volume of a 0.1W0.3N aqueous solution of sodium hydroxide. The extracted liquid is neutralized, acidic ingredients are removed and the supernatant dialysis liquid is dried to obtain an alkali-extracted polysaccharide. The polysaccharide is added with an alkaline aqueous solution and phenol, stirred to obtain a homogeneous solution, added with an acidic substance and left resting to separate the mixture into two solution phases and an intermediate precipitate phase. The objective polysaccharides composed of 3 kinds of recurring units of formula IWformula III can be separated from the upper phase. The number of the unit of formula III of the polysaccharide is 4W12 per 100 recurring units and that of the unit of formula II and formula III is 16W30 each.
