PURPOSE: To provide the title compsn. containing a novel protease having an improved stability, and good storage and heat stability.
CONSTITUTION: The objective compsn. contains stabilized protease, in which a naturally occurring amino acid residue other than proline is substituted with a proline residue at one or more positions, at which position(s) the dihedral angles and ψ constitute values within the intervals of -90°<<-40° and -180°<ψ<180° preferably -90°<<40° and 120°<ψ<180° or -90°<<40° and -50°<ψ<10°, and which position(s) are not located in regions, in which the protease is characterized by possessing α-helical or '-sheet structure. The above mentioned protease may be obtained by analyzing the secondary structure of the protease and subsequently substituting the naturally occurring amino acid residue of the specific region of the gene encoding the protease with the proline residue by the induction of mutation and subsequently developing the gene encoding the stabilized protease in the host organism.
