Abstract:
PURPOSE: To obtain a pharmaceutical composition capable of releasing a drug at a controlled rate over a long period by mixing a polyester-type biodegradable polymer with a natural or synthetic peptide salt in dried state, finely pulverizing the mixture, preliminarily pressing and heating the powder and extruding the product. £ GH-RH, calcitonin, embedding material, endless screw. CONSTITUTION: A biodegradable polymer selected from poly-1,4-butylene succinate, poly-2,3-butylene succinate, poly-1,4-butylene fumarate and poly-2,3- butylene fumarate is mixed with an active component consisting of the pamoate, tannic acid salt, stearic acid salt or palmitic acid salt of a natural or synthetic peptide in dried state until the average size of the produced powder reaches about <500μm. The obtained mixture is preliminarily pressed and heated and then extruded at 90-100 deg.C under a pressure of 50-500kg/cm<2> , the extruded material is cooled and optionally finely pulverized at a low temperature and the powder is classified. The above peptide is e.g. LH-RH and somatostatin.
