Abstract:
The invention relates to a method of producing a randomly primed cDNA suitable for targeted cloning, which is based on the fact that during the synthesis of the first cDNA strand a single-strand oligonucleotide mixture is attached to an RNA or an (m)RNA mixture obtained from a sample. Said oligonucleotide mixture is characterized in that the 3' component comprises random sequences, preferably between 4 and 10 nucleotides in length, and the 5' component presents a recognition site for a restriction endonuclease when it is transferred to a double strand. The recognition site is preferably a recognition site for a restriction endonuclease which generates protruding ends.
