Abstract:
PROBLEM TO BE SOLVED: To provide a method for obtaining a heterodimer immunoglobulin or a fragment thereof by genetic engineering and a method for making the same.SOLUTION: According to the present invention, there is provided a heterodimeric immunoglobulin or a fragment thereof comprising a first genetically engineered immunoglobulin chain and a second genetically engineered immunoglobulin chain, each comprising a genetic engineered domain with a protein-protein interface. Here, at least one amino acid residue at the protein-protein interface of the parent domains of the first genetically engineered immunoglobulin chain and the second genetically engineered immunoglobulin chain is replaced with an amino acid residue from the protein-protein interface of the donor domain of the first member of the naturally occurring immunoglobulin superfamily at the same position on 3D structure.SELECTED DRAWING: Figure 1
