Abstract:
PROBLEM TO BE SOLVED: To provide monoclonal antibody-substituting binding molecules that specifically bind to a target of interest with high affinity and specificity.SOLUTION: Disclosed herein is a bispecific Fn3-based binding molecule comprising a first non-Fn3 binding sequence and a second non-Fn3 binding sequence, where, in the first non-Fn3 binding sequence at least one amino acid is altered in at least one of the bottom AB, CD or EF loop regions or the C terminal of the first non-Fn3 binding sequence compared to the wild type Fn3 domain, and the first non-Fn3 binding sequence binds to a frist target, and in the second non-Fn3 binding sequence, at least one amino acid is altered in at least one of the top BC, DE, or FG loop regions of the Fn3 domain compared to the wild type Fn3 domain, and the second non-Fn3 binding sequence binds to a second target where the first and second targets are present on different molecules.SELECTED DRAWING: None
