Abstract:

To provide a method for producing a fine particle of a narrow particle size distribution containing a hydrophilic physiologically active substance.

The method for producing a fine particle containing a hydrophilic physiologically active substance comprises (a) a step of forming a reversed phase emulsion by mixing an aqueous solvent having dissolved therein a hydrophilic physiologically active substance with a water-immiscible organic solvent having dissolved therein an amphiphatic polymer composed of a hydrophobic segment of a poly(hydroxy acid) and a hydrophilic segment of a polysaccharide or polyethylene glycol, (b) a step of obtaining solid matter containing the hydrophilic physiologically active substance by removing the solvents from the reversed phase emulsion, (c) a step of dispersing the solid matter containing the hydrophilic physiologically active substance in a solvent A in which the hydrophobic segment of the amphiphatic polymer is soluble while the hydrophilic segment of the amphiphatic polymer is insoluble, and (d) a step of adding the solid matter containing the dispersion of the hydrophilic physiologically active substance obtained in the step (c) to a solvent B which has dissolved therein a surface-modifier and is miscible with the solvent A and in which the hydrophobic segment of the amphiphatic polymer is insoluble.
