Abstract:

PURPOSE: To obtain the title anchor for the high-yield, high-efficiency separation and purification of DNA-bound proteins by putting a functional group capable of binding to a solid carrier at one end of nucleic acid fragment followed by binding the frictional group, to the solid carrier activated.

CONSTITUTION: A functional group (e.g. sulfhydryl group) capable of binding to a desired solid carrier is put at one end of nucleic acid fragment, and poly dT sequence is put at the other end to bind a nucleic acid probe having poly A strand to this poly dT sequence, and the resulting probe is brought into contact with the solid carrier activated with a functional group (e.g. maleimide group) reactive with the former functional group to effect binding the nucleic acid to said solid carrier by the reaction between these functional groups, thus obtaining the objective nucleic acid-immobilized anchor. When the former functional group is an aliphatic primary amino group, the nucleic acid can be bound to the solid carrier by a reaction following activation of said carrier with succinymisyl group.
