Abstract:

PURPOSE: To obtain an antibacterial substance (asperatin) having high safety in high efficiency by culturing a koji-mold in a medium composed of protease- treated steamed cereals by static culture and purifying the obtained antibacterial substance by the treatment with an organic solvent and an agent capable of fractionating into two layers.
CONSTITUTION: This antibacterial substance (asperatin) having high safety and exterminating cultured cancer cell in vitro is produced by screening a brewing koji-mold 238 strain belonging to the genus Aspergillus, selecting Aspergillus oryzae A0K260 having antibacterial activity to beer yeast, culturing the strain in a medium composed of protease-treated steamed cereals (e.g. steamed rice) at 30°C for 2 weeks by static culture, filtering the culture liquid, recovering the filtrate, treating the recovered filtrate with equal amount of carbon tetrachloride for 30 min to effect the denaturation of protein, subjecting the product to centrifugal separation, collecting the supernatant, dissolving 1M Nail and 20% polyethylene glycol in the supernatant, washing the precipitate with ethanol and freeze-drying the product.
