Abstract:
To provide a composition and a method for inducing the modification of a target endogenous nucleic acid sequence in the nucleus of a eukaryotic cell.SOLUTION: The present invention provides a composition for inducing the modification of a target endogenous nucleic acid sequence in the nucleus of a eukaryotic cell, including a Cas9/RNA complex. The complex includes recombinant Cas9 protein, and a guide RNA including crRNA and tracrRNA. The Cas9/RNA complex is a combination of the recombinant Cas9 protein and the guide RNA. The combination is assembled through the in vitro mixing and incubation of the recombinant Cas9 protein with the guide RNA. The guide RNA is either transcribed in vitro or synthesized chemically. The target endogenous nucleic acid sequence includes a part complementary to the crRNA of the guide RNA. The combination of the recombinant Cas9 protein and the guide RNA is introduced into a eukaryotic cell, causing modification of the target endogenous nucleic acid sequence in the nucleus of the eukaryotic cell.SELECTED DRAWING: Figure 1a
